About Mazdutide
Mazdutide (development codes IBI362 and LY3305677) is a once-weekly dual GLP-1 and glucagon receptor agonist. Clinical programme status, as reported in the published record: a randomised, placebo-controlled multiple-ascending-dose phase 1b study in Chinese adults with overweight or obesity was published in EClinicalMedicine in 2021 (Ji et al.). A randomised, double-blind, placebo-controlled phase 2 trial was published in Nature Communications in 2023 (Ji et al.). The phase 3 GLORY-1 trial, "Once-Weekly Mazdutide in Chinese Adults with Obesity or Overweight", was published in the New England Journal of Medicine in 2025 (Ji et al., doi:10.1056/NEJMoa2411528). In June 2025 mazdutide received approval from China's National Medical Products Administration for chronic weight management. Mazdutide is not approved in the United Kingdom, the European Union or the United States. Peptx supplies mazdutide only as lyophilised reference material for laboratory research. Mechanism
Mazdutide is a synthetic analogue based on the mammalian oxyntomodulin scaffold, engineered for once-weekly exposure. It binds and activates two class B G-protein-coupled receptors in parallel: the glucagon-like peptide-1 receptor (GLP-1R) and the glucagon receptor (GCGR). This distinguishes it from single-receptor GLP-1 agonists such as semaglutide and from the GLP-1/GIP co-agonist tirzepatide, which recruits the glucose-dependent insulinotropic polypeptide receptor instead of GCGR. Published pharmacology describes it as a "glucagon-like peptide-1 (GLP-1) and glucagon receptor dual agonist" (Ji et al., Nature Communications, 2023). Receptor-level research interest centres on how parallel GLP-1R and GCGR signalling differs from GLP-1R-only or GLP-1R/GIPR signalling in in-vitro and preclinical systems.
