    Mechanism

    GLP-1 and GIP are the two principal incretin hormones. Their receptors, GLP-1R and GIPR, are class B G-protein-coupled receptors that couple to Gs and adenylate cyclase, and both are expressed in pancreatic islets as well as in central and peripheral tissues. A dual agonist is a single molecule engineered to recruit both receptors rather than one.

    VK2735 is described in the peer-reviewed literature as a GLP-1/GIP receptor dual agonist. Mechanistically it therefore sits in the same receptor pairing as tirzepatide, and in a different pairing from oxyntomodulin-based GLP-1/glucagon dual agonists such as mazdutide, or from triple agonists that add glucagon receptor activity.

    Viking Therapeutics has reported development of both a subcutaneous and an oral formulation. Formulation route affects exposure and absorption characteristics; it does not change the receptor targets.

    Clinical programme status

    • Investigational. VK2735 is not approved in the United Kingdom, European Union or United States.
    • Phase 2 subcutaneous data from the VENTURE study were published in the peer-reviewed journal Obesity in 2026.
